Abstract
The invention relates to a new method of characterising a target polynucleotide. The method uses a pore and a Dda helicase. The helicase controls the movement of the target polynucleotide through the pore. The invention also relates to modified Dda helicases which can be used to control the movement of polynucleotides and are particularly useful for sequencing polynucleotides.
Claims19
A DNA-dependent ATPase (Dda) helicase in which at least one cysteine residue and/or at least one non-natural amino acid have been introduced into (i) the tower domain and/or (ii) the pin domain and/or (iii) the 1A (RecA-like motor) domain, wherein the helicase retains its ability to control the movement of a polynucleotide.

A Dda helicase which comprises a variant of SEQ ID NO:
- 8 comprising (a) A360K;
- (b) Y92L and/or A360Y;
- (c) Y92L, Y350N and Y363N;
- (d) Y92L and/or Y363N or (e) Y92L, wherein the helicase retains its ability to control the movement of a polynucleotide.
