Abstract
Methods for identifying cancer patients amenable to anti-cancer immunotherapy are provided along with methods of monitoring cancer therapy. Also provided are methods of treating cancer patients amenable to anti-cancer immunotherapy. The methods involve determining the level of CD127<low>PD-1<low>T cells. The patients are treated with an immune checkpoint inhibitor, such as an anti-CTLA-4 antibody, e.g. ipilimumab.
Claims53
A method of identifying a human cancer subject amenable to an anti-cancer immunotherapy, comprising:
- (a) obtaining a dataset comprising data representing the level of CD127low PD-1low T cells in the human cancer subject;
- (b) obtaining a dataset comprising data representing the level of CD127low PD-1low T cells in a control, wherein the control is the human cancer patient at an earlier time, a distinct human cancer subject that is unresponsive to the anti-cancer immunotherapy, or a healthy human;
- and
- (c) assessing the amenability of the human cancer subject to the anti-cancer immunotherapy based on the data, optionally, identifying the human cancer subject as amenable to anti-cancer immunotherapy when the level of CD127low PD-1low T cells is greater in the human cancer subject than the level of CCD127low PD-1low T cells in the control.
A method of monitoring anti-cancer immunotherapy in a human cancer subject comprising:
- (a) obtaining a dataset comprising data representing the level of CD127low PD-1low T cells in the human cancer subject;
- (b) obtaining a dataset comprising data representing the level of CD127low PD-1low flow T cells in a control, wherein the control is the human cancer subject at an earlier time, a distinct human cancer subject that is unresponsive to anti-cancer immunotherapy, or a healthy human, wherein a greater level of CD127low PD-1low T cells in the human cancer subject relative to control is indicative of a response to the anti-cancer immunotherapy;
- and/or a lower level of CD127low PD-1low T cells in the human cancer subject relative to control is indicative of a lack of response to the anti-cancer immunotherapy.
A method of treating cancer in a human cancer subject comprising:
- administering a therapeutically effective amount of a composition comprising an immune checkpoint inhibitor to the subject, wherein the human cancer subject has been determined to have a greater level of CD127low PD-1low cells than a control, wherein the control is the human cancer subject at an earlier time, a distinct human cancer subject that is unresponsive to anti-cancer immunotherapy, or a healthy human, thereby treating the cancer in the human cancer subject, optionally further comprising administering a soluble growth factor to the human cancer subject.
A composition comprising:
- (a) a sample from a human cancer subject comprising T cells;
- (b) an anti-CD127 antibody;
- and (c) an anti-PD-1 antibody.
