Compositions, Devices, Systems, and Methods for Using a Nanopore

A method, comprising:

• (a) providing a device comprising:
• a substrate having a pore therein, the substrate separating a first fluid chamber from a second fluid chamber;
• and
• a power source electrically coupled to electrodes, wherein the power source and electrodes are adapted to apply a potential difference between the first fluid chamber and the second fluid chamber;
• (b) introducing macromolecules to the first fluid chamber;
• (c) applying a potential difference between the first fluid chamber and the second fluid chamber such that a macromolecule is drawn toward the second fluid chamber, to position the macromolecule at the pore;
• (d) maintaining the macromolecule at the pore for a period of time;
• and
• (e) after the period of time, repeating steps (c) and (d) one or more times to position one or more additional macromolecules at the pore in series.
