Abstract
The present invention relates to methods of repairing tissue. More specifically, the present invention relates to methods of using cells and an implantable support for the repair of tissue defects, where the implantable support and cells are implanted into the tissue defect less than 2 hours after the cells are applied to the support.
Claims11
An in vivo method of repairing connective tissue in a mammalian_subject in need thereof comprising the steps of:
- (a) providing an implantable support comprising a collagen-containing membrane and a sample of autologous chondrocyte cells;
- (b) heating the implantable support to between 35° C. and 37° C.;
- (c) applying said sample of cells to the surface of said support to produce an implantable matrix and incubating said implantable matrix for between 15 minutes to 1 hour and 50 minutes such that at least 90% of said cells adhere to the surface of the matrix;
- and
- (d) implanting said matrix into or adjacent to said connective tissue in need of repair.

An in vivo method of repairing connective tissue in a mammalian subject in need thereof comprising the steps of:
- (a) isolating chondrocyte cells from said mammalian subject;
- (b) culturing said isolated chondrocyte cells to produce cultured chondrocyte cells;
- (c) harvesting said cultured chondrocyte cells to provide a sample of about 106 cultured chondrocyte cells suspended in said subject's own serum;
- (d) providing an implantable support comprising a collagen-containing membrane;
- (e) heating the implantable support to between 35° C. and 3° C.;
- (f) applying said sample of about 106 cultured chondrocyte cells to the surface of said support to produce an implantable matrix and incubating said implantable matrix for between 15 minutes to 1 hour and 50 minutes such that at least 90% of said chondrocyte cells adhere to the surface of the matrix;
- and
- (g) implanting said matrix into or adjacent to said connective tissue in need of repair.
