Certain chemical entities, compositions, and methods

A method of treating a disorder mediated by Raf in a subject suffering from the disorder, the method comprising administering to the subject a therapeutically effective amount of a compound of Formula II:

• or a pharmaceutically acceptable salt thereof, wherein
• R1, R6, R7, R8, and R10 are independently hydrogen, cyano, halo, hydroxy, azido, nitro, carboxy, sulfinyl, sulfanyl, sulfonyl, optionally substituted alkoxy, optionally substituted cycloalkyloxy, optionally substituted aryloxy, optionally substituted heteroaryloxy, optionally substituted heterocycloalkyloxy, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted alkenyl, optionally substituted aryloxy, optionally substituted aryl, optionally substituted heteroaryl, optionally substituted heterocycloalkyl, optionally substituted amino, optionally substituted acyl, optionally substituted alkoxycarbonyl, optionally substituted aminocarbonyl, optionally substituted aminosulfonyl, optionally substituted carbamimidoyl, or optionally substituted alkynyl;
• R2, R3, R4, and R5 are independently hydrogen, halo, cyano, or optionally substituted alkyl;
• R9 is hydrogen, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, or optionally substituted heteroaryl;
• and
• R11 is hydroxyl, formyl, optionally substituted alkoxy, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl, —COR12, —CO2R12, —CONR12R13, —C(NR14)NR12R13, —C(NCN)NR12R13, —SO2NR12, or —SO2NR12R13, where R12 is optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, or optionally substituted heteroaryl;
• and R13 and R14 are independently hydrogen, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, or optionally substituted heteroaryl;
• or R12 and R13 may be joined together with any intervening atoms to form an optionally substituted heterocycloalkyl ring;
• or R9 and R11 may be joined together with any intervening atoms to form an optionally substituted heterocycloalkyl ring.

A method of treating a disorder mediated by Raf in a subject suffering from the disorder, the method comprising:

• a. determining the presence or absence of a BRAF mutation in a biological sample isolated from the subject;
• and
• b. if a BRAF mutation is determined to be present in the subject, administering to the subject a therapeutically effective amount of a compound of Formula II:
• or a pharmaceutically acceptable salt thereof, wherein
• R1, R6, R7, R8, and R10 are independently hydrogen, cyano, halo, hydroxy, azido, nitro, carboxy, sulfinyl, sulfanyl, sulfonyl, optionally substituted alkoxy, optionally substituted cycloalkyloxy, optionally substituted aryloxy, optionally substituted heteroaryloxy, optionally substituted heterocycloalkyloxy, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted alkenyl, optionally substituted aryloxy, optionally substituted aryl, optionally substituted heteroaryl, optionally substituted heterocycloalkyl, optionally substituted amino, optionally substituted acyl, optionally substituted alkoxycarbonyl, optionally substituted aminocarbonyl, optionally substituted aminosulfonyl, optionally substituted carbamimidoyl, or optionally substituted alkynyl;
• R2, R3, R4, and R5 are independently hydrogen, halo, cyano, or optionally substituted alkyl;
• R9 is hydrogen, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, or optionally substituted heteroaryl;
• and
• R11 is hydroxyl, formyl, optionally substituted alkoxy, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl, —COR12, —CO2R12, —CONR12R13, —C(NR14)NR12R13, —C(NCN)NR12R13, —SO2NR12, or —SO2NR12R13, where R12 is optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, or optionally substituted heteroaryl;
• and R13 and R14 are independently hydrogen, optionally substituted alkyl, optionally substituted cycloalkyl, optionally substituted heterocycloalkyl, optionally substituted aryl, or optionally substituted heteroaryl;
• or R12 and R13 may be joined together with any intervening atoms to form an optionally substituted heterocycloalkyl ring;
• or R9 and R11 may be joined together with any intervening atoms to form an optionally substituted heterocycloalkyl ring.
