Abstract
A resolution enhancement technique for a line scanning confocal microscopy system that generates vertical and horizontal line scanning patterns onto a sample is disclosed. The line scanning confocal microscopy system is capable of producing line scanning patterns through the use of two alternative pathways that generate either the vertical line scanning pattern or horizontal line scanning pattern.
Claims21
A line scanning microscopy system (100) comprising:
- a light source (102) for transmitting a single light beam (101);
- a scanning apparatus arrangement (114,116,124,126) for relaying the single light (101) beam along either a first pathway (A) for generating line scans (172) that form a vertical line-scan pattern (170) or a second pathway (B) for generating line scans (182) that form a horizontal line-scan pattern (180);
- an optic arrangement (112, 132) for relaying the line scans (172) of the vertical line-scan pattern (170) or line scans (182) of the horizontal line-scan pattern (180) to illuminate a sample (134) and generate fluorescent line scan emissions (160) formed in either the vertical line-scan pattern (170) or the horizontal line-scan pattern (180);
- a scaling component for scaling the fluorescent line scan emissions (172, 182) to either locally contract each of the fluorescent line scan emissions (172, 182) to produce a contracted fluorescent line scan emissions (172′, 182′) that form a contracted line-scan pattern (170′, 180′), or expand each of the fluorescent line-scan emissions (172, 182) to produce an expanded fluorescent line scan emissions (172″, 182″) to form an expanded line-scan pattern (170″, 180″);
- and
- a detection apparatus (148) for capturing an image of the expanded line-scan pattern (170″, 180″) or the contracted line-scan pattern (170′, 180′).
A microscopy system (200) comprising:
- a first light source (203) that transmits a first single light beam (215) and a second light source (201) that transmits a second single light beam (217);
- a first optic arrangement (207) that combines the first single light beam (215) and the second single light beam (217) to form a combined light beam (219);
- a scanning apparatus arrangement (210) for relaying the combined light beam (219) along either a first pathway (A) for generating line scans that form a vertical line-scan pattern (170) or a second pathway (B) for generating line scans that form a horizontal line-scan pattern (180);
- a second optic arrangement (212) for relaying the line scans of the vertical line-scan pattern (170) or line scans of the horizontal line-scan pattern (180) to illuminate a sample (234) and generate a fluorescent line-scan emissions (260) formed in either the vertical line-scan pattern (180) or the horizontal line-scan pattern (180);
- a scaling component for scaling the fluorescent line scan emissions (260) to either locally contract each of the fluorescent line scan emissions (260) to produce a contracted fluorescent line scan emissions (172′, 182′) to form a contracted line-scan pattern (170′, 180′), or expand each of the fluorescent line-scan emissions (260) to produce an expanded fluorescent line scan emissions (172″, 182″) to form an expanded line-scan pattern (170″, 180″);
- and
- a detection apparatus (170″, 180″) for capturing an image of the expanded line-scan pattern (170″, 180″) or the contracted line-scan pattern (170′, 180′).
A microscopy system (300) comprising:
- a light source (302) for transmitting a single light beam (304);
- a scanning apparatus arrangement (310, 316) for relaying the single light beam (304) and generating a light sheet (304A);
- an excitation objective (322) for imaging the light sheet (304A) onto a sample (324) for generating fluorescence emissions (305);
- a detection objective (326) for relaying the fluorescence emissions (305) through an optic arrangement;
- a scaling component for scaling the fluorescence emissions (305)received from the optic arrangement to either locally contract the fluorescence emissions (305)arising from the light sheet to produce fluorescence emissions (305)that are contracted or locally expand the fluorescence emissions (305)arising from the light sheet to produce fluorescence emissions (305)that are expanded;
- and
- a detection apparatus (338)for capturing an image of the expanded or the contracted fluorescence emissions (305)arising from the light sheet.
