Abstract
Substance productivity is improved by introducing a metabolic pathway for synthesis of acetyl-CoA or acetic acid from glucose-6-phosphate into yeast. Acetic acid productivity, acetyl-CoA productivity, and productivity of a substance made from acetyl-CoA-derived are improved by attenuating genes involved in the glycolytic system of yeast and introducing a phosphoketolase gene into the yeast.
Claims2
A recombinant, non-xylose fermenting Saccharomyces cerevisiae, which comprises an attenuated phosphofructokinase gene endogenous to said Saccharomyces cerevisiae, an introduced phosphoketolase gene, an introduced Clostridium acetobutylicum acetoacetic acid decarboxylase gene, an introduced Clostridium acetobutylicum butyrate-acetoacetate CoA-transferase subunit A gene, an introduced Clostridium acetobutylicum butyrate-acetoacetate CoA-transferase subunit B gene, an introduced Clostridium acetobutylicum acetyl-CoA acetyltransferase gene, and an introduced Clostridium acetobutylicum isopropanol dehydrogenase gene, wherein isopropanol production by said Saccharomyces cerevisiae is enhanced compared with wild-type strain, and wherein said introduced phosphoketolase gene encodes the amino acid sequence of any one of SEQ ID NOS:
- 1 to 19.
