Abstract
The present invention provides a method for providing modified mRNAs of reduced immunogenicity and/or immunostimulatory capacity for use in protein replacement therapy. The invention further provides modified mRNAs and pharmaceutical compositions comprising the modified mRNAs according to the invention for use in protein replacement therapy.
Claims21
Method for targeted modulation, preferably reduction, of the immune response against an mRNA coding for at least one biologically active polypeptide or protein, wherein the method comprises the steps of
- (1) identifying a target mRNA wild type sequence coding for the biologically active polypeptide or protein and
- (2) modifying at least 70% of the codons of the wild type sequence which are cytosine content optimizable by increasing the cytosine-content of the mRNA such that the cytosine-content of the region of the coding region of the modified mRNA is larger than the cytosine-content of the coding region of the wild type mRNA, whereby the encoded amino acid sequence is unchanged compared to the wild type sequence and, optionally
- (3) modifying at least 70% of the codons for amino acids which are not eligible for C-optimization, but are guanosine-content optimizable by increasing the guanosine-content
