Abstract
The present invention relates to diagnostic, prognostic and therapeutic methods related to anxiety, as well as related products. In particular examples the anxiety is post-traumatic stress disorder.
Claims33
A method for diagnosing an anxiety disorder in a female subject, comprising:
- a. collecting a tissue sample from the subject;
- b. analyzing pituitary adenylate cyclase-activating polypeptide (PACAP) protein expression levels in the tissue sample;
- and
- c. diagnosing the anxiety disorder in the subject having elevated tissue PACAP expression levels as compared to a control tissue sample obtained from a subject who does not have an anxiety disorder.
A method for identifying a subject at risk of developing an anxiety disorder, comprising:
- a. collecting a DNA sample from the subject;
- b. analyzing the DNA sample for the presence of a SNP in a PAC1 receptor gene;
- and
- c. diagnosing the anxiety disorder in the subject having the SNP in the PAC1 receptor gene.
A method for identifying a subject at risk of developing an anxiety disorder, comprising:
- a. collecting a DNA sample from the subject;
- b. analyzing the DNA sample for epigenetic methylation at a first site within a ADCYAP1R1 CpG island;
- and
- c. diagnosing the anxiety disorder in the subject having elevated levels of epigenetic methylation.
A method of treating an anxiety disorder in a female subject in need thereof, comprising administering to the subject a therapeutically effective amount of a PACAP inhibitor.
A DNA amplification kit for diagnosing an anxiety disorder in a female subject, comprising:
- a. two or more oligonucleotide primers capable of binding to DNA sequence flanking position rs2267735 of the ADCYAP1R1 gene;
- b. optimization buffer;
- c. polymerase;
- and
- d. deoxyribonucleotide triphophates (dNTPs);
- wherein each of elements a-d is arranged in one or more containers.
An immunoassay kit for diagnosing an anxiety disorder in a female subject, comprising:
- a. a primary IgG anti-PACAP antibody;
- b. an anti-IgG immuno-conjugated secondary antibody;
- c. diluent and wash buffers;
- d. chromagen;
- and
- e. pre-coated microplate;
- wherein each of elements a-e is arranged in one or more containers.
A gene chip comprising a solid surface, having attached thereto a plurality of oligonucleotides, wherein the oligonucleotides have a sequence complementary to a portion of a PAC1 receptor gene or a flanking sequence thereof, wherein at least some of the portions of the PAC1 receptor gene have a SNP.
A composition comprising an 8-9 amino acid peptide of SEQ ID NO:3 or SEQ ID NO:4.
An antibody that binds specifically to an epitope at the carboxy-terminal end of PACAP.
A composition comprising an antibody that binds specifically to a peptide of SEQ ID NO:3 or SEQ ID NO:4.
