APP SPECIFIC BACE INHIBITORS (ASBIs) AND USES THEREOF

A method of ameliorating one or more symptoms of Alzheimer's disease, and/or reversing Alzheimer's disease, and/or reducing the rate of progression of Alzheimer's disease, said method comprising:

• administering to a subject in need thereof an APP specific BACE inhibitor (ASBI) in an amount sufficient to ameliorate one or more symptoms of Alzheimer's disease, and/or to reverse Alzheimer's disease, and/or to reduce the rate of progression of Alzheimer's disease, wherein said APP specific BACE inhibitor is a flavonoid characterized by the formula:
• wherein:
• R1 is selected from the group consisting of OH, and O-saccaharide;
• R4 and R5 are independently selected from the group consisting of H, OH, NH2, O-alkyl, O-trifluoromethyl, S-alkyl, S-aryl, carboxylate, halogen, NH-alkyl, N,N-dialkyl, NHCO-alkyl, and heteroaryl, alkyl urea, and carbamate;
• and
• R2 and R3 are independently selected from the group consisting of H, OH, NH2, O-alkyl, O-trifluoromethyl, S-alkyl, S-aryl, carboxylate, halogen, NH-alkyl, N,N-dialkyl, NHCO-alkyl, heteroaryl, alkyl urea, and carbamate.

A method of slowing the progression, stopping, or reversing age-related macular degeneration (AMD) in a mammal, said method comprising administering to said mammal an APP specific BACE inhibitor, wherein said inhibitor is a flavonoid characterized by the formula:

• R1 is selected from the group consisting of OH, O-saccaharide, O-alkyl, O-trifluoromethyl, O-aryl, O-heteroaryl;
• R4 and R5 are independently selected from the group consisting of H, OH, NH2, O-alkyl, O-trifluoromethyl, S-alkyl, S-aryl, carboxylate, halogen, NH-alkyl, N,N-dialkyl, NHCO-alkyl, and heteroaryl, alkyl urea, and carbamate;
• and
• R2 and R3 are independently selected from the group consisting of H, OH, NH2, O-alkyl, O-trifluoromethyl, S-alkyl, S-aryl, carboxylate, halogen, NH-alkyl, N,N-dialkyl, NHCO-alkyl, heteroaryl, alkyl urea, and carbamate;
• and/or
• an ASBI prodrug that comprises a galangin prodrug.
