Compounds, compositions, methods of making, and methods of using analogues of (−)-picropodophyllin, as well as a transgenic animal model and its use for identifying anticancer agents.
Claims19
A compound of formula I:
- wherein X, X′, Y, Y′ and Z are independently H, OCH3, OCH2CH3, O-i-CH2CH2CH3, O-n-CH2CH2CH3, SCH3, SCH2CH3, S-i-CH2CH2CH3, S-n-CH2CH2CH3, S(O)CH3, S(O)CH2CH3, S(O)-i-CH2CH2CH3, S(O)-n-CH2CH2CH3, S(O)2CH3, S(O)2CH2CH3, S(O)2-i-CH2CH2CH3, S(O)2-n-CH2CH2CH3, CN, SCN, OCN, N3, halogen, CH3, CH2CH3, i-CH2CH2CH3, n-CH2CH2CH3, CF3, CCH, CCCH3, CH═CH2, Ph, NH2, NHCH3, N(CH3)2, NHCH2CH3, N(CH2CH3)2, NO, NO2, CHO, COCH3, COCH2CH3, CO2H, CO2CH3, CO2CH2CH3, OC(O)H, OC(O)CH3, OSO3H, OSO3CH3, OSO3CH2CH3, OPO3H2, OPO3HCH3, OPO3HCH2CH3, OPO3(CH3)2, OPO3(CH2CH3)2, P(O)(OH)2, P(O)(OCH3)(OH), P(O)(OCH2CH3)(OH), P(O)(OCH3)2, or P(O)(OCH2CH3)2 with the proviso that when X and X′ are H, Y, Y′, and Z cannot all be methoxy and that when X′ is H, and Y, Y′, and Z are methoxy, X cannot be a halogen.
A compound of formula II:
- wherein X, X′, Y, Y′ and Z are independently hydrogen, a C1-C8 alkyl, or C1-C8 alkoxy group with the proviso that when X and X′ are H, Y, Y′, and Z cannot all be OCH3;
- and
- R1 may be oxo, —OH, —OCH3, —OC2H5, —OC3H7, —OC4H9, —OCH2CH═CH2, —OCH2Ph, —OCH2CH2NH2, —OCOH, —OCOCH3, —OCH2OH, —OC2H4OH, —OC3H6OH, —OC4H8OH, or a glycoside.
