Abstract
An anti-Ang2 antibody or an antigen-binding fragment thereof that specifically binds to an angiogenesis-inducing factor Angiopoietin-2 (Ang2) and complexes with a Tie2 receptor and Ang2, and related methods and compositions.
Claims24
An anti-Ang2 antibody or an antigen-binding fragment thereof which specifically binds to Angiopoietin-2 (Ang2) and forms a complex with a Tie2 receptor and Ang2.
A hybridoma cell line of accession number KCLRF-BP-00295.
A method for screening a candidate drug for preventing or treating a disease related to Ang2 overexpression, angiogenesis, increase of vascular permeability, or a decrease in normal blood vessel formation, comprising
- a) contacting a candidate compound to a specimen containing Ang2 and a Tie2 receptor;
- and
- b) identifying i) the formation of the complex in which the candidate compound, Ang2, and the Tie2 receptor are bound, ii) Tie2 receptor activation, or both of Ang2 inhibition and Tie2 receptor activation, or iii) both of i) and ii),
- wherein identification of (i) or (ii), or both, indicates the candidate compound is a candidate drug for preventing or treating a disease related to Ang2 overexpression, angiogenesis, vascular permeability enhancement, or a decrease in normal blood vessel formation.
A method for screening for anti-Ang2 antibodies comprising:
- contacting a candidate compound to an epitope of Ang2 having the amino acid sequence of SEQ ID NO:
- 11 in the presence of a Tie2 receptor;
- measuring binding affinity between the epitope and the candidate compound;
- and
- identifying binding between Ang2 and a Tie2 receptor,
- wherein the epitope of Ang2 is Q418, P419, a combination of Q418 and P419, or 2 to 20 amino acid residues comprising the Q418, P419 or the combination of Q418 and P419, in Ang2 of SEQ ID NO:
- 11;
- and wherein the candidate compound is determined as an anti-Ang2 antibody when the binding between the epitope and the candidate compound is detected.
A polypeptide molecule comprising:
- at least one selected from the group consisting of a polypeptide comprising the amino acid sequence of SEQ ID NO:
- 1, a polypeptide comprising the amino acid sequence of SEQ ID NO:
- 2, and a polypeptide comprising the amino acid sequence of SEQ ID NO:
- 3;
- at least one selected from the group consisting of a polypeptide comprising the amino acid sequence of SEQ ID NO:
- 4, a polypeptide comprising the amino acid sequence of SEQ ID NO:
- 5, and a polypeptide comprising the amino acid sequence of SEQ ID NO:
- 6;
- or
- a combination thereof.
