Abstract
The present invention relates to isolated monoclonal anti-EGFR antibodies, and to the use of such antibodies and antibody fragments to detect EGFR, particularly those expressed by cancer cells. The antibodies are useful in diagnostic as well as therapeutic indications. Methods, devices and kits for the immunodetection and immunotherapy of cells expressing EGFR is also encompassed.
Claims50
An isolated antibody specific for epithelial growth factor receptor (EFGR), comprising a light chain variable domain comprising at least one complementarity determining region (CDR) selected from SEQ ID NOs:3, SEQ ID NO:4, and SEQ ID NO:5, and a heavy chain variable domain comprising at least one CDR selected from SEQ ID NOs:6, SEQ ID NO:7, and SEQ ID NO:8.
An isolated antibody specific for EFGR, comprising a light chain variable domain comprising three CDRs comprising SEQ ID NO:3, SEQ ID NO:4, and SEQ ID NO:5, and a heavy chain variable domain comprising three CDRs comprising SEQ ID NO:6, SEQ ID NO:7, and SEQ ID NO:8.
An isolated antibody specific for EFGR, comprising a light chain variable domain comprising:
- a) a CDR1 comprising SEQ ID NO:3,
- b) a CDR2 comprising SEQ ID NO:4, and
- c) a CDR3 comprising SEQ ID NO:5;
- and
- a heavy chain variable domain comprising
- a) a CDR1 comprising SEQ ID NO:6,
- b) a CDR2 comprising SEQ ID NO:7,
- c) a CDR3 comprising SEQ ID NO:8.
The antibody of any preceding claim, wherein the antibody is capable of binding to different forms of EGFR selected from the group consisting of:
- a soluble form;
- a membrane-bound form;
- a phosphorylated form;
- a non-phosphorylated form;
- a homodimer form;
- and a heterodimer form.
The antibody of any preceding claim, wherein the antibody is capable of binding to EGFR with an affinity of between 10−8 and 10−11 M.
The antibody of any preceding claim, wherein the antibody is humanized.
The antibody of any preceding claim, wherein the antibody is capable of selectively reducing the activity of EGFR.
The antibody of any preceding claim, wherein the antibody is labeled.
The antibody of claim 13, wherein the antibody is labeled fluorescently, with an enzyme, or with a radioisotope.
The antibody of any preceding claim bound to EGFR.
The antibody of any preceding claim bound to EGFR and further bound to a solid support.
A pharmaceutical composition comprising the antibody of any preceding claim and a pharmaceutically acceptable carrier and/or diluent.
The antibody of claim 47, wherein the light chain and the heavy chain are at least 80%, at least 85%, at least 90%, at least 95%, or at least 98% identical to the light chain and the heavy chain of the antibody of any of claims 1-16.
An isolated nucleic acid encoding an antibody of any of claims 1-16 and 47-48.
A host cell comprising the nucleic acid of claim 49.
A cell line expressing the antibody of any of claims 1-16 and 47-48.
The cell line of claim 51, wherein the cell line is a hybridoma.
A method of producing an antibody of any of claims 1-16 and 47-48, comprising culturing the host cell of claim 50 in culture medium under conditions sufficient to produce the antibody.
