Abstract
Molecular markers are described for determining the presence or absence of a gene conferring resistance to tomato yellow leaf curl virus from S. chilense (Ty-1) in a host plant. Also described, are methods for producing a host plant comprising a gene conferring resistance to tomato yellow leaf curl virus from S. chilense (Ty-1), including the analysis of the presence or absence of the molecular markers. A plant, and parts thereof, obtained by such a method are also described.
Claims18
A molecular marker for determining presence or absence of a gene conferring resistance to tomato yellow leaf curl virus (TYLCV) from S. chilense (Ty-1) in a host plant, said host plant resulting from introgression of Ty-1 into a TYLCV-susceptible plant, whereby the marker is located between markers Msc09983-6 and M005H10 on chromosome 6.
A marker for determining presence or absence of a gene conferring resistance to tomato yellow leaf curl virus (TYLCV) from S. chilense (Ty-1) in a host plant, wherein the marker is a nucleic acid marker specific for the Ty-1 gene or the protein encoded by said gene, wherein the marker comprises any of the SNPs and indels specified in the sequence depicted in FIG. 7.
A method for producing a host plant comprising a gene conferring resistance to tomato yellow leaf curl virus from S. chilense (Ty-1), the method comprising the steps of:
- (a) introgressing genetic material from S. chilense into a TYLCV-susceptible plant to produce a host plant that comprises genetic material from S. chilense;
- (b) analyzing said host plant for the presence of the Ty-1 resistance allele with a marker that is located between markers Msc09983-6 and M005H10 on chromosome 6;
- and
- (c) selecting one or more host plants comprising said resistance allele of S. chilense.
A method for producing a host plant comprising a nucleotide sequence conferring resistance to tomato yellow leaf curl virus, the method comprising the steps of:
- (a) optionally providing a transformation vector comprising a nucleotide sequence encoding the protein encoded by the sequence of the Ty-1 gene;
- (b) transforming a plant susceptible for tomato yellow leaf curl virus with the vector of step (a) or direct with a nucleotide sequence encoding the protein encoded by the sequence of the Ty-1 gene;
- and
- (c) selecting transformants which express said nucleotide sequence.
A transgenic plant harboring a heterologous nucleotide sequence capable of producing a protein that is encoded by the sequence of the Ty-1 gene as depicted in FIG. 7.
