Abstract
The invention describes new peptides containing epitopes recognized by CD4+ natural killer T (NKT) cells for increasing activity for use in infectious diseases, autoimmune diseases, immune reaction to administration of allofactors, allergic diseases, therapy of tumors, prevention of graft rejection and prevention of immunization against viral proteins used in gene therapy or gene vaccination.

An isolated immunogenic peptide derived from an antigenic protein comprising:
- (1) a natural killer T (NKT) cell epitope of said antigenic protein comprising a [FWHY]-xx-[ILMV]-xx-[FWHY] motif;
- and
- (2) a thioreductase [CST]-xx-C or C-xx-[CST] motif, which is either immediately adjacent to said NKT cell epitope, or separated from said NKT cell epitope by a linker of at most 7 amino acids.
A method for preparing a peptide capable of eliciting NKT cell activation, said method comprising the steps of:
- (1) providing a peptide sequence of an antigenic protein comprising a NKT cell epitope comprising a [FVVHY]-xx-[ILMV]-xx-[FWHY] motif, and
- (2) linking to said peptide sequence a sequence comprising a thioreductase [CST]-xx-C or C-xx-[CST] motif, such that said oxidoreductase motif and said NKT cell epitope are either immediately adjacent to each other or separated by a linker of at most 7 amino acids.
An in vitro method for obtaining a population of antigen-specific NKT cells, the method comprising the steps of:
- providing peripheral blood cells;
- contacting said cells in vitro with an immunogenic peptide comprising (1) a NKT cell epitope of an antigenic protein comprising a [FWHY]-xx-[ILMV]-xx-[FWHY] motif and (2) an oxidoreductase C-XX-[CST] or [CST]-XX-C motif which is either immediately adjacent to said NKT cell epitope, or separated from said NKT cell epitope by a linker of at most 7 amino acids;
- and
- expanding said cells in the presence of IL-2, IL-15 or IL-7.
A method for obtaining a population of antigen-specific NKT cells, the method comprising the steps of:
- providing an immunogenic peptide comprising (1) a NKT cell epitope of an antigenic protein comprising a [FWHY]-xx-[ILMV]-xx-[FWHY] motif and (2) an oxidoreductase C-XX-[CST] or [CST]-XX-C motif, which is either immediately adjacent to said NKT cell epitope, or separated from said NKT cell epitope by a linker of at most 7 amino acids;
- administering said immunogenic peptide to a subject, and
- obtaining said population of said antigen-specific NKT cells from said subject.
