Abstract
Methods for producing recombinant cell populations are disclosed. The disclosed methods may be used to produce therapeutic polyclonal proteins.
Claims30
A method for producing a cell population, said method comprising the steps of:
- a) transforming a population of host cells with at least one vector comprising at least one copy of a nucleic acid sequence encoding a protein of interest that integrates into the genomic DNA of a subset of the population of host cells thereby generating a population of host cells capable of producing the protein of interest,
- wherein the population of host cells comprises transformed host cells and non-transformed host cells and wherein said generation of the population of host cells capable of producing the protein of interest does not require clonal selection of an individual progenitor cell clone;
- and
- b) maintaining the population of host cells to produce the protein of interest.

A method for producing a polyclonal cell population, said method comprising the steps of:
- a) transforming one or more host cells with at least one vector comprising at least one copy of a nucleic acid sequence encoding a component of a polyclonal protein composition using a rep protein or a rep-like protein, wherein the rep protein or rep-like protein mediates integration of the nucleic acid sequence into the host cell genome and said transformation does not require clonal selection of an individual cell;
- b) generating a cell population capable of producing the component of the polyclonal protein composition;
- c) repeating steps (a)-(b) at least once to generate a cell population capable of producing a different component of the polyclonal protein composition;
- and
- d) mixing at least two cell populations generated by steps (a)-(c) to produce a polyclonal cell population capable of producing the polyclonal protein composition.
