Abstract
The present invention relates to anti-drug vaccines based on conjugates between the drug and a non-immunogenic carrier protein. In preferred embodiments, it provides for anti-cocaine vaccines and their use to diminish the effects and/or use of cocaine in a subject.

A pharmaceutical composition comprising a drug/Self protein conjugate, in an amount effective at inducing an immune response to the drug in an immunocompetent subject, together with a suitable pharmaceutical carrier, where the Self protein is not immunogenic in said subject.
A pharmaceutical composition comprising a conjugate between a Self protein and a hapten selected from the group consisting of cocaine, a cocaine derivative, and a transition state analog of cocaine, in an amount effective at inducing an immune response to cocaine in an immunocompetent subject, together with a suitable pharmaceutical carrier, where the Self protein is not immunogenic in said subject.
The pharmaceutical composition of claim 12 where the Self protein is human serum albumin.
The pharmaceutical composition of claim 12 where the Self protein is a human immunoglobulin.
The pharmaceutical composition of claim 10 which further comprises an adjuvant.
A method of treating a subject in need of such treatment comprising administering to the subject a pharmaceutical composition comprising a drug/Self protein conjugate, in an amount effective at inducing an immune response to the drug in the subject, together with a suitable pharmaceutical carrier, where the Self protein is not immunogenic in said subject.
A method of treating a subject in need of such treatment comprising administering to the subject a pharmaceutical composition comprising a conjugate between a Self protein and a hapten selected from the group consisting of cocaine, a cocaine derivative, and a transition state analog of cocaine, in an amount effective at inducing an immune response to cocaine in an immunocompetent subject, together with a suitable pharmaceutical carrier, where the Self protein is not immunogenic in said subject.
A cocaine pre-hapten having Formula II:
- wherein R1 and R2 are the same or different, and may be:
- unsubstituted or substituted, branched or unbranched, alkyl where one or more substituent may be hydroxyl (OH) or a ketone (═O);
- or
- unsubstituted or substituted, branched or unbranched, alkoxy (preferably C1-C4 or C1-C2 alkoxy), where one or more substituent may be OH or a ketone.
