Abstract
Processes for synthesizing aminosulfone compounds are provided. Aminosulfone compounds obtained using methods provided herein are useful in production or synthesis of isoindoline based PDE 4 modulators.

A process for preparing a compound of Formula (I):
- or a salt, solvate, or stereoisomer thereof, wherein R1 and R2 are each independently hydrogen, substituted or unsubstituted (C1-C6)alkyl, substituted or unsubstituted (C1-C6)alkoxy, (C3-C18)cycloalkyl, cyano, or (C3-C18)cycloalkyl-(C1-C6)alkoxy,
- comprising the steps of:
- (1) reacting a nitrile of Formula (III):
- with LiCH2SO2CH3, to form an enamine of Formula (IV):
- and
- (2) reducing the enamine of Formula (IV) to form the compound of Formula (I) or a salt, solvate, or polymorph thereof.
