Abstract
The present invention relates to the treatment, diagnosis, and prophylaxis of cancer based on the expression of foxp3.
Claims28
An isolated nucleic acid molecule selected from the group consisting of:
- (a) complements of nucleic acid molecules that hybridize under high stringency conditions to a second nucleic acid molecule comprising a nucleotide sequence set forth as SEQ ID NO:6, foxp3 (SEQ ID NO:1) lacking exons 2 and 3, or foxp3 exons 1 and 4 joined together,
- (b) nucleic acid molecules that differ from the nucleic acid molecules of (a) in codon sequence due to the degeneracy of the genetic code, and
- (c) full-length complements of (a) or (b);
- or
- a nucleotide sequence that is at least about 90% identical to a nucleotide sequence set forth as SEQ ID NO:6, foxp3 (SEQ ID NO:1) lacking exons 2 and 3, or foxp3 exons 1 and 4 joined together, or a full-length complement thereof.

A siRNA molecule that selectively targets SEQ ID NO:6, foxp3 (SEQ ID NO:1) lacking exons 2 and 3, or a nucleic acid molecule comprising foxp3 exons 1 and 4 joined together.
A method for diagnosing cancer in a subject comprising
- obtaining a biological sample from the subject, and
- determining the expression in the biological sample of a FoxP3 polypeptide or a nucleic acid molecule that encodes the FoxP3 polypeptide, wherein the nucleic acid molecule comprises SEQ ID NO:1, or a nucleotide sequence that is at least 90% identical to the nucleotide sequence of SEQ ID NO:1,
- wherein the cancer is not pancreatic carcinoma and
- wherein the expression in the biological sample of the polypeptide or the nucleic acid molecule that encodes it is indicative of the subject having cancer,
- optionally wherein the biological sample is free of regulatory T cells (Treg;
- CD4+CD25+FOXP3+).

A method for diagnosing cancer in a subject comprising
- obtaining a biological sample from the subject, and
- determining the expression in the biological sample of a FoxP3 polypeptide or a nucleic acid molecule that encodes the FoxP3 polypeptide, wherein the nucleic acid molecule comprises (1) SEQ ID NO:6, foxp3 (SEQ ID NO:1) lacking exons 2 and 3, or a nucleic acid molecule comprising foxp3 exons 1 and 4 joined together, or (2) a nucleotide sequence that is at least 90% identical to the nucleotide sequence of (1),
- wherein the expression in the biological sample of the polypeptide or the nucleic acid molecule that encodes it is indicative of the subject having cancer,
- optionally wherein the biological sample is free of regulatory T cells (Treg;
- CD4+CD25+FOXP3+).
