Abstract
The present invention relates to the genes involved in the biosynthetic pathway for sesquiterpenes of SB type (alpha-santalene, epi-beta-santalene, cis-alpha-bergamotene, trans-alpha-bergamotene and endo-beta-bergamotene,) and for the precursor thereof, Z,Z-farnesyl diphosphate (Z,Z-FPP), involving a Z,Z-FPP synthase and a sesquiterpene SB synthase, and to the uses thereof for producing sesquiterpeniques compounds of SB type.

A method for producing sesquiterpenes in a cell comprising:
- a) introducing into said cell a construct having an expression cassette comprising a first gene encoding a Z,Z-farnesyl diphosphate synthase (zFPS) having at least 80% identity to SEQ ID NO:
- 2 and a construct having an expression cassette comprising a second gene encoding a SB synthase (SBS) having at least 80% identity to SEQ ID NO:
- 4;
- and
- b) culturing the transformed cell in the presence of isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP) under conditions suitable for the expression of said first and said second genes.
An isolated or recombinant protein comprising:
- a) an amino acid sequence with at least 80% identity to SEQ ID NO:
- 2 and having Z,Z-FPP synthase activity;
- b) SEQ ID NO:
- 2;
- or
- c) an amino acid sequence having at least 80% identity to SEQ ID NO:
- 4 and SB synthase activity.
A method for identifying a genomic polymorphism between S. habrochaites and species of the Solanum type that are sexually compatible with S. habrochaites comprising detecting a genomic polymorphism with molecular markers comprising all or part of a nucleic acid with at least 80% identity to SEQ ID NO:
- 1 or SEQ ID NO:
- 3 so as to control the introgression of the corresponding genes in said species.
A method comprising introducing a zFPS gene encoding a zFPS having at least 80% identity to SEQ ID NO:
- 2 and a SBS gene encoding a SB synthase having at least 80% identity to SEQ ID NO:
- 4 in a species not sexually compatible with Solanum habrochaites by protoplast fusion.
