Abstract
The invention relates to compositions and methods of using electrophoresis separation matrices. The invention provides nano-particle comprising separation matrices having increased conductivity at low voltage.
Claims28
A gel composition comprising a nano-particle formed from a conductive material, wherein the size of the nano-particle ranges from about 1 nm to about 30 nm.
A composition comprising:
- a. a separation matrix;
- b. buffer;
- and,
- c. a nano-particle hydrosol, wherein the nano-particle sizes range from about 1 to about 30 nm.
A composition comprising:
- a. a separation matrix, wherein the separation matrix is selected from the group consisting of agarose, polyacrylamide, hydroxyalkeylcellulose, polyvinyl alcohol, and mixtures thereof;
- b. buffer, wherein the buffer is a tris-based buffer;
- and,
- c. a nano-particle hydrosol, wherein the nano-particle is formed from a metal selected from the group consisting of copper, ruthenium, rhodium, palladium, silver, rhenium, osmium, iridium, platinum, gold, mercury, and mixtures thereof.
A method of making a conductive matrix for capillary electrophoresis comprising:
- a. mixing a nano-particle dispersion in a buffer resulting in a nano-buffer suspension;
- b. mixing separation matrix powder in the nano-buffer suspension;
- and,
- c. heating the separation matrix-nano-buffer suspension until the matrix is melted.
A method of separating nucleic acids comprising:
- a. mixing a nano-particle dispersion in a buffer resulting in a nano-buffer suspension;
- b. mixing separation matrix powder in the nano-buffer suspension;
- c. heating the separation matrix-nano-buffer suspension until the matrix is melted and gelled;
- d. loading nucleic acid samples into the gelled matrix;
- and,
- e. electrophoresing the nucleic acid samples into the gelled matrix at a low voltage.
