Abstract
Lavendamycin analogues, methods for their synthesis, and methods for their use in the treatment of diseases such as cancer and HIV infection are described.
Claims21
A compound having a formula:
- or a pharmaceutically acceptable salt thereof, wherein:
- X is hydrogen or R10(O)—;
- Y is hydroxy, alkoxy, amino, Ph(CH2)mO—, HO(CH2)nO— or —O(CH2)oOPO3H2, wherein m, n, and o are each independently 1, 2, 3, 4, 5, 6, 7, 8, 9 or 10;
- R1 is hydrogen, amino, alkoxy, halo, acyl, aziridinyl, pyrrolidinyl, piperidinyl or pyridinyl;
- R2, R3, R4, R5, R6, R7, and R8 are each independently hydrogen or a substituted or unsubstituted radical selected from the group consisting of alkyl, aryl, cycloalkyl, alkenyl, alkynyl, heteroalkyl, heterocyclic, heteroalkenyl, heteroalkynyl, halo, nitro, cyano, alkoxy, amino, amido, thioamido, acyl, thioacyl, and imino;
- and
- R10 is hydrogen or a substituted or unsubstituted radical selected from the group consisting of alkyl, aryl, cycloalkyl, alkenyl, alkynyl, heteroalkyl, heterocyclic, heteroalkenyl, and heteroalkynyl;
- with a proviso that when R1 is hydrogen, halo, amino, alkoxy or acyl, then Y is not hydroxy, alkoxy or amino.

A method of synthesizing a compound having a formula:
- wherein:
- X is hydrogen or R10C(O)—;
- Y is hydroxy, alkoxy or amino;
- Ra and Rb are either (a) independently hydrogen or a substituted or unsubstituted radical selected from the group consisting of alkyl, aryl, cycloalkyl, alkenyl, alkynyl, heteroalkyl, heterocyclic, heteroalkenyl, and heteroalkynyl or (b) together with the nitrogen atom to which they are attached form a three-, four-, five-, six-, seven- or eight-membered cyclic ring;
- R1 is amino, aziridinyl, pyrrolidinyl, piperidinyl or pyridinyl;
- R2, R3, R4, R5, R6, R7, and R8 are each independently hydrogen or a substituted or unsubstituted radical selected from the group consisting of alkyl, aryl, cycloalkyl, alkenyl, alkynyl, heteroalkyl, heterocyclic, heteroalkenyl, heteroalkynyl, halo, nitro, cyano, alkoxy, amino, amido, thioamido, acyl, thioacyl, and imino;
- and
- R10 is hydrogen or a substituted or unsubstituted radical selected from the group consisting of alkyl, aryl, cycloalkyl, alkenyl, alkynyl, heteroalkyl, heterocyclic, heteroalkenyl, and heteroalkynyl;
- the method comprising:
