Abstract
A hybrid type I polyketide synthase gene typically containing a starter module and a plurality of heterologous extender modules is used to synthesise novel polyketides. It is preferably under the control of type II polypolyketide synthase promoter e.g. act I or S.coelicolor.
Claims32
A hybrid polyketide synthase (“PKS”) gene comprising a first nucleic acid portion or portions encoding at least one domain of a first type I PKS and a second nucleic acid portion or portions encoding at least one type I PKS domain which is heterologous to said first PKS.
3-23. (canceled)
A method for producing a gene assembly encoding a hybrid type I polyketide synthase (PKS), said method comprising joining:
- a) a first nucleic acid molecule which encodes at least one domain of a first type I PKS, and
- b) a second nucleic acid molecule which encodes at least one domain of a second type I PKS ;
- thereby forming a gene assembly encoding a functional hybrid type I PKS.
The method of clam 40, wherein the second nucleic acid molecule additionally encodes a thioesterase domain.
A method for producing a gene assembly encoding a hybrid type I polyketide synthase (PKS), said method comprising replacing, in a gene assembly encoding a first type I PKS, a first nucleic acid molecule encoding at least one domain with a second nucleic acid molecule encoding said at least one domain, wherein said second nucleic acid molecule is from a second type I PKS.
The method of 45, wherein said first nucleic acid molecule encodes an AT domain.
The method of 47, wherein said second nucleic acid molecule encodes for an AT domain, wherein the AT domain is selected from the group consisting of the AT domains of the PKSs for erythromycin, rapamycin, avermectin, monensin, tetronasin, oleandomycin, amphotericin and rifamycin.
The method of 48, wherein said second nucleic acid molecule encodes for an AT domain from an extension module.
The method of 48, wherein said second nucleic acid molecule encodes for an AT domain from the rapamycin PKS.
The method of 45, wherein said gene assembly encoding a first type I PKS encodes the erythromycin PKS.
The method of 47, wherein the AT domain encoded for by said first nucleic acid molecule has a different specificity than the AT domain encoded for by said second nucleic acid molecule.
