Abstract
Multi-armed functional polyethylene glycol polymers with active terminal groups may be used in conjugation with small molecule pharmaceuticals, proteins and peptides to enhance half-life and bioavailability of the drug and improve solubility, stability and immunological competence.
Claims25
A multi-armed polyethylene glycol, comprising general formula I:
- RPEGzCOX I
- wherein, R is a straight-chain alkane containing 1 to 10 carbon atoms, isopropyl or benzyl group;
- z is an integer representing the number of polyethylene glycol arms;
- and X is a functional group.
A multi-branched, functional polyethylene glycol polymer comprising at least one functional moiety, a plurality of individual polyethylene glycol arms linked to the functional moiety by a carboxyl group and a terminal moiety selected from the group consisting of straight chain alkanes having from 1 to 10 carbon atoms, isopropyl or benzyl group
A method of making a multiple branched functional polyethylene polymer having the general formula RPEGzCOX, wherein wherein, R is a straight-chain alkane containing 1 to 10 carbon atoms, isopropyl or benzyl group;
- z is an integer representing the number of polyethylene glycol arms;
- and X is a functional group, comprising the steps of:
- a. reacting a first R terminated polyethylene glycol with a functional small molecular moiety to yield a first functionalized polyethylene glycol;
- and
- b. reacting the first functionalized polyethylene glycol with at least one tri-functional molecular moiety;
- c. reacting a second R terminated polyethylene glycol with a functional small molecular moiety to yield a second functionalized polyethylene glycol;
- and
- d. reacting the first functionalized polyethylene glycol arm with the second functionalized polyethylene glycol.
