Abstract
A bioartificial liver system is described that incorporates a cell reservoir and hepatocyte spheroids to both increase the number of and longevity of cells in the system. Additional methods are also described for forming spheroid aggregates from isolated hepatocytes.
Claims20
A system for treating a biological fluid from a mammal, said system comprising:
- a) a bioreactor comprising a selectively permeable barrier separating a fluid treatment compartment and a cell compartment;
- b) a cell reservoir in fluid communication with said cell compartment of said bioreactor;
- and
- c) a rocking device in contact with said cell reservoir to induce motion in fluid contained in said cell reservoir.
1 more element
A method for treating a biological fluid from a mammal, said method comprising:
- a) providing a system comprising:
- i) a bioreactor comprising a selectively permeable barrier separating a fluid treatment compartment and a cell compartment;
- ii) a cell reservoir in fluid communication with said cell compartment of said bioreactor;
- iii) a rocking device in contact with said cell reservoir;
- and
- iv) a cell fluid comprising a plurality of cells for treating said blood, wherein said cell fluid is in fluid communication with said cell compartment and said cell reservoir;
- b) removing said biological fluid from said mammal;
- c) introducing said biological fluid into said fluid treatment compartment of said bioreactor;
- and
- d) allowing said biological fluid to flow through and exit said fluid treatment compartment, thereby treating said biological fluid.
7 more elements
A method for treating blood from a mammal, said method comprising:
- a) providing a system comprising
- i) a bioreactor comprising a selectively permeable barrier separating a fluid treatment compartment and a cell compartment;
- ii) a cell reservoir in fluid communication with said cell compartment of said bioreactor;
- iii) a rocking device in contact with said cell reservoir;
- and
- iv) a cell fluid comprising a plurality of cells for treating said blood wherein said cell fluid is in fluid communication with said cell compartment and said cell reservoir;
- b) placing said fluid treatment compartment in fluid communication with the bloodstream of said mammal;
- and
- c) allowing said blood from said mammal to flow through and exit said fluid treatment compartment, thereby treating said blood.
6 more elements
A method of forming spheroid aggregates, said method comprising:
- a) providing a reservoir having a plurality of hepatocytes in a cell medium;
- and
- b) rocking said plurality of hepatocytes at a frequency and for a duration sufficient for spheroid aggregates to form.
