Abstract
A method and a system for separating glycated hemoglobin from non-glycated hemoglobin, and a method for one-read quantitative determination of the glycated hemoglobin and %HbA1c that does not require an additional measurement of total hemoglobin.
Claims3
1:
- A method for directly measuring the concentration of glycated hemoglobin and the percent hemoglobin A1c in blood which comprises the steps of diluting and lysing a sample of said blood with an aqueous buffer to provide a diluted and lysed blood sample buffered in the range of 7.5 to about 10;
- providing an immobilized boronic acid derivative formed by covalent conjugation of a boronic acid compound to a solid substrate;
- intimately contacting said immobilized boronic acid with said diluted, lysed, and buffered blood solution for the formation of the cyclic ester between the dihydroxy boryl group of the immobilized boronic acid and the 1,2-cis-diol group of the carbohydrate moiety of the HbA1c molecule in said blood sample;
- separating the esterified mobilized boronic acid substrate and the remainder of said blood solution from each other;
- mixing the separated, immobilized boronic acid substrate with a developing solution for the development of a dye compound;
- and measuring the intensity of dye so formed.

2:
- A kit adapted for the determination of the percentage of HbA1c in the total hemoglobin of a blood sample comprising:
- (a) a boronic acid immobilized onto a solid support;
- (b) a lysing composition for a blood sample to be analyzed;
- (c) a buffering composition for buffering a diluted solution of a blood sample to be analyzed to a pH in the range of about 7.5 to about 10;
- and (d) a developing solution reactive with the cyclic ester formed between dihydroxy boral group of a boronic acid derivative and the 1,2-cis-diol group in the carbohydrate moiety of an HbA1c molecule to form a colored compound.
2 more elements
3:
- A kit in accordance with claim 2 wherein said kit additionally includes a lysing accelerator.
