Abstract
Fluorescent protein expressing cells with very high fluorescence are described as well as a method to obtain such cells. The fluorescence of such cells can be as high as about 1000 times the auto-fluorescence of the respective host cell. Due to the high fluorescence, said cells are very suitable tools in the development, surveillance and/or optimization of production processes, for selection of culture media and process parameters and for the determination of cell growth and the detection, quantification and monitoring of biomass.
Claims22
Mammalian cells genetically modified with a fluorescent protein expressing gene, wherein the signal of the fluorescent protein is at least 100 times higher than the auto-fluorescence of the cells.
A process for obtaining mammalian cells genetically modified with a fluorescent protein expressing gene, preferably GFP, or better still, humanized GFP, wherein the cells are transfected thus, that at least 25% transient transfection is observed after 24 hours, preferably about 30% or more, and much preferably about 50% or more, and that the most active genetically modified clone(s) is (are) selected.
