The inventions relate to compositions and articles of manufacture comprising connexin modulators, pannexin modulators, gap junction modulators, hemichannel modulators, and pannexin channel modulators and their use, alone or in combination, in treating ocular and other disorders.

A method for treating a subject for a neurodegenerative disease or disorder, comprising administering a therapeutically effective amount of a connexin hemichannel modulator to said subject, wherein the connexin hemichannel modulator is the compound N-(6-Acetyl-3-hydroxy-2,2-dimethyl-3,4-dihydro-2H-chromen-4-yl)-3-chloro-4-fluorobenzamide (tonabersat).
