A preparation method of Auricularia auricula polysaccharide, comprising the steps of:
- S1) drying and crushing Auricularia auricula, and then degreasing the Auricularia auricula by a supercritical fluid extraction process to obtain a degreased Auricularia auricula;
- wherein in S1, conditions of the supercritical fluid extraction process are as follows:
- a flow rate of CO2 is in a range of 7-12 L/h, a pressure is in a range of 12-25 MPa, a temperature is in a range of 45-60° C., and an extraction time is in a range of 1-2 h;
- S2) adding the degreased Auricularia auricula obtained in S1 into water, adding snailase, performing preliminary enzymolysis, deactivating the snailase, concentrating and drying an obtained system to obtain a preliminary enzymolysis product;
- S3) adding the preliminary enzymolysis product prepared in S2 into a H2O2 solution, carrying out ultrasonic treatment, adding sodium bisulfite thereto, carrying out alcohol precipitation, centrifuging and drying to obtain an Auricularia auricula raw sugar extract;
- S4) dissolving the Auricularia auricula raw sugar extract prepared in S3 in water, adding a complex enzyme thereto, conducting the secondar enzymolysis, and deactivating the complex enzyme to obtain a secondary enzymolysis extract, wherein the complex enzyme is at least one selected from the group consisting of β-glucanase, saccharifying enzyme, cellulase, pectinase, α-amylase, and α-glucosidase;
- S5) respectively streaking Lactobacillus bulgaricus, Streptococcus thermophilus and Bifidobacterium longum in a Gauze's medium, and carrying out activation culture to obtain a strain seed liquid;
- S6) inoculating the strain seed liquid prepared in S5 into the secondary enzymolysis extract prepared in S4, and carrying out fermentation culture, followed by concentration, alcohol precipitation and centrifugation to obtain a fermented Auricularia auricula polysaccharide;
- S7) adding the fermented Auricularia auricula polysaccharide prepared in S6 into water, adding sodium sulfate and a phosphorylation reagent thereto, adjusting a pH value of an obtained mixture to 8.8-9.2, heating and subjecting the resulting mixture to reaction, dialyzing, and concentrating to obtain a phosphorylated Auricularia auricula polysaccharide solution;
- S8) adding the phosphorylated Auricularia auricula polysaccharide solution obtained in S7 into a Sevag reagent, subjecting the resultant mixture to reaction under stirring, centrifuging to deproteinize the phosphorylated Auricularia auricula polysaccharide solution,
- repeating the preceding deproteinization steps on the resulting liquid 1-3 times and removing the remaining Sevag reagent therein under reduced pressure to obtain a deproteinized Auricularia auricula polysaccharide;
- S9) adding activated carbon and the deproteinized Auricularia auricula polysaccharide prepared in S8 into water, performing adsorption under stirring, filtering, performing alcohol precipitation on the resulting liquid, and centrifuging to obtain a refined Auricularia auricula polysaccharide;
- S10) dissolving the refined Auricularia auricula polysaccharide prepared in S9 and trisodium citrate in water, adding a zinc salt, adjusting a pH value of the resulting solution to 7.2-7.5, heating and subjecting the resulted mixture to reaction under stirring, filtering, performing alcohol precipitation on the resulting liquid, centrifuging, collecting the resulting solid, and freeze-drying the resulting solid collected to obtain the Auricularia auricula polysaccharide.
