Abstract
Disclosed herein are peptide sequences capable of directing adeno-associated viruses (AAV) to target specific environments, for example the nervous system and the heart, in a subject. Also disclosed are AAVs having non-naturally occurring capsid proteins comprising the disclosed peptide sequences, and methods of using the AAVs to treat diseases.
Claims18
A method of delivering a nucleic acid to a nervous system of a subject in need, comprising:
- providing a composition comprising an adeno-associated virus (AAV), wherein the AAV comprises a capsid protein that comprises an AAV peptide comprising the amino acid sequence of DGTLAVPFKAQ (SEQ ID NO:
- 4), and wherein the AAV comprises a nucleic acid to be delivered to the nervous system of the subject;
- and
- administering the composition to the subject,
- wherein the AAV capsid protein comprises the amino acid sequence of SEQ ID NO:
- 4 inserted between positions corresponding to residues 586-591 of an AAV capsid protein sequence of SEQ ID NO:
- 45.
