Abstract
A polypeptide containing an antibody Fc region variant which has an amino acid sequence in which an amino acid alteration at position 238 according to EU numbering is combined with other specific amino acid alteration(s), was found to have decreased binding activities to all activating FcγRs, in particular FcγRIIa (R type), while maintaining its FcγRIIb-binding activity, when compared to a polypeptide containing a native IgG Fc region.
Claims16
A method of producing a polypeptide comprising an IgG Fc region variant, the method comprising:
- preparing a host cell comprising a nucleic acid encoding the polypeptide comprising the IgG Fc region variant;
- expressing the nucleic acid in the host cell, thereby producing the polypeptide;
- and
- collecting the produced polypeptide,
- wherein the amino acid sequence of the IgG Fc region variant comprises one of the following combinations of amino acid residues at the indicated EU numbering positions:
- (i) 235F/238D/241M/296E,
- (ii) 235F/238D/241M/268P/296E/324H,
- (iii) 235F/237Q/238D/241M/296E/322A,
- (iv) 235F/237Q/238D/241M/296E/322E,
- (v) 235F/237Q/238D/241M/268P/296E/322A,
- (vi) 235F/237Q/238D/241M/268P/296E/322E,
- (vii) 235F/237Q/238D/241M/268P/296E/322A/324H, or
- (viii) 235F/237Q/238D/241M/268P/296E/322E/324H.

- (iii) 238D/241M/268P/296E,
- (iv) 238D/241M/296E/324H,
- (v) 238D/241M/296E/330H, or
- (vi) 238D/241M/296E/330Y.
