The present disclosure relates, according to some embodiments, to systems, apparatus, compositions, methods, and workflows that include DNase I variants with desirable properties including, for example, salt tolerance. A DNase I variant, in some embodiments, may have an amino acid sequence that is at least 85% identical, at least 90% identical, at least 95% identical, and/or at least 98% identical to SEQ ID NO:1 and may be identical to SEQ ID NO:1 at one or more positions selected from the group of positions corresponding to L29, A35, D87, Q88, S94, P103, T108, P121, P132, A135, D145, E161, G172, P190, H208, and A224 of SEQ ID NO:1.
A DNase I variant having an amino acid sequence that is (a) at least 95% identical to SEQ ID NO:1 and (b) identical to SEQ ID NO:1 at one or more positions corresponding to L29, A35, D87, Q88, S94, P103, T108, P121, P132, A135, D145, E161, G172, P190, H208, and A224 of the polypeptide of SEQ ID NO:1, wherein said variant has DNase I activity.
A DNase I variant having an amino acid sequence (I) that is at least 95% identical to SEQ ID NO:1 and (II) that comprises one or more of:
A DNase I variant having an amino acid sequence that is at least 98% identical to SEQ ID NO:1, wherein said variant has DNase I activity.
A DNase I variant comprising an amino acid sequence identical to the amino acid sequence of SEQ ID NO:
A DNase I variant having an amino acid sequence having at least 93% identity to SEQ ID NO:3 and comprising one or more substitutions at positions corresponding to SEQ ID NO:3, the substitutions and corresponding positions selected from N18A, S43K, S43R, S103X, N106A, S206K, S206R, T207K, and T207R, wherein said variant has DNase I activity.
