Abstract
The invention provides a method of differentiating human pluripotent stem cells to ectodermal cell by treating human pluripotent stem cells, which are dissociated single cells, and inducing differentiation to ectodermal cells under conditions where a ROCK(Rho-kinase) inhibitor is present in a culture medium in contact with the cells after dissociation of the human pluripotent stem cells.
Claims16
A method of improving the differentiation efficacy of human pluripotent stem cells to ectodermal cells, which comprises culturing human pluripotent stem cells, which are dissociated single cells, in a serum-free culture medium which is suitable for induction of differentiation of pluripotent stem cells into ectodermal cells by a Serum-free Floating culture of Embryoid Body-like aggregates (SFEB) method,
- wherein the culture medium comprises a Rho kinase (ROCK) inhibitor for at least 2 days,
- wherein, during culturing, induction of differentiation occurs, and
- wherein the ectodermal cells are neural cells, or neural precursor cells.
