Abstract
Disclosed herein is a composition including a recombinant nucleic acid sequence that encodes an antibody or fragment thereof that targets an immune checkpoint molecule. The disclosure also provides a method of preventing and/or treating disease in a subject using said composition and method of generation.
Claims16
A composition for generating a synthetic antibody in a subject comprising one or more nucleic acid molecules encoding one or more antibodies or fragments thereof, wherein the one or more antibodies or fragments target at least one immune checkpoint molecule selected from the group consisting of PD-1, LAG-3, PD-L1, GITR, CD40, OX40, CTLA-4, TIM-3, 4-1BB, and a combination thereof,
- wherein the one or more nucleic acid molecules comprises a nucleotide sequence encoding an amino acid sequence having at least about 80% identity over the entire length of an amino acid sequence selected from the group consisting of SEQ ID NOs:
- 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26, and 28, wherein said amino acid sequence comprises heavy and light chain CDR amino acid sequences selected from the group consisting of SEQ ID Nos:
- 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26, and 28.
A composition for generating a synthetic antibody in a subject comprising one or more nucleic acid molecules encoding one or more antibodies or fragments thereof, wherein the one or more antibodies or fragments target at least one immune checkpoint molecule selected from the group consisting of PD-1, LAG-3, PD-L1, GITR, CD40, OX40, CTLA-4, TIM-3, 4-1BB, and a combination thereof,
- wherein the one or more nucleic acid molecules comprises a nucleotide sequence encoding an amino acid sequence having at least about 90% identity over the entire length of at least one amino acid sequence selected from the group consisting of SEQ ID NOs:
- 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26, and 28, wherein said amino acid sequence comprises the heavy and light chain CDRs of an amino acid sequence selected from the group consisting of SEQ ID Nos:
- 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26, and 28.
