Abstract
The present invention includes a low dose and sustained release formulation of a mitochondrial uncoupling agent The compositions of the invention are useful for preventing or treating a disease or disorder, such as non-alcoholic fatty liver disease, non-alcoholic steatohepatitis, insulin resistance and/or diabetes, in a subject in need thereof.
Claims9
A pharmaceutical composition in unit dose form comprising:
- a mitochondrial uncoupling agent and at least one pharmaceutically acceptable carrier,
- wherein the pharmaceutical composition is formulated for oral administration;
- wherein the mitochondrial uncoupling agent is coated with an extended release coating comprising at least one water soluble polymer and at least one water insoluble polymer, and
- wherein oral administration of the pharmaceutical composition to a subject provides a steady state plasma concentration of about 0.05 μM to about 200 μM of the mitochondrial uncoupling agent in the subject without toxic effect.

A pharmaceutical composition in unit dose form comprising:
- 2,4-dinitrophenol (2,4-DNP) and at least one pharmaceutically acceptable carrier,
- wherein the pharmaceutical composition is formulated for oral administration;
- wherein the DNP is coated with an extended release coating comprising at least one water soluble polymer and at least one water insoluble polymer, and
- wherein oral administration of the pharmaceutical composition to a subject provides a steady state plasma concentration of about 0.05 μM to about 200 μM of the DNP in the subject without toxic effect.
