Abstract
One embodiment provides a commensal gut production platform for ex vivo production of human gut commensal microbiota. Another embodiment provides devices, systems and methods for ex vivo culturing of gut microflora in a system that mimics the human gut environment. The culturing of the commensal microbiota in the disclosed systems produces gut microbiota having defined characteristics and properties that can be exploited to treat various conditions in a subject.
Claims15
A method comprising:
- screening a first sample of gut microbiota obtained from a subject to detect and remove ova, parasites, or viruses;
- determining a first set of relative proportions and identities of the microbiota in the first sample;
- culturing the first sample to expand specific populations of microbiota in the presence of a spatial gradient of dissolved oxygen;
- and
- storing the expanded culture from the first sample, wherein the expanded culture comprises 20%-25% butyrate producers, 15-20% short chain fatty acid producers, 10-12% mucin degrading organisms, 1-5% mucin producers, 10-15% digestive enzyme producers, 0.5-1.0% neuro-endocrine peptide producers, 2-5% secondary bile acid metabolizers, 1-2% polyphenol and isothiocyanate producers, 5% tight junction protein production inducers, and 0.5-4% histone deacetylase producers and DNA methylase producers.
