Abstract
The present invention relates to use of DHA analogs and their pharmaceutical compositions for treating ototoxicity, by administering these compounds or pharmaceutical compositions to subjects in need thereof. Ototoxicity is defined as damage to the structures of the ear, such as the cochlea and vestibular system, by drugs or toxins. Ototoxicity can result in irreversible hearing loss, tinnitus, dysequilibrium, Meniere's disease, or vertigo.
Claims14
A method for treating ototoxicity comprising administering to a subject in need thereof a therapeutically effective amount of a compound of Formula I, II, III or IV:
- wherein
- represents a cis or trans bond;
- R1 is —C(O)ORa, —C(O)NRbRc, —C(O)H, —C(NH)NRbRc, —C(S)H, —C(S)ORa, —C(S)NRbRc, or —CN;
- R2 and R3 are each independently H or a protecting group;
- R4 and R5 are each independently H, halo, unsubstituted or substituted C1-C6 alkyl, unsubstituted or substituted C3-C10 carbocycle, unsubstituted or substituted heterocycle comprising one or two 3-, 4-, 5- or 6-member rings and 1-4 heteroatoms selected from N, O and S,
- Ra is a protecting group or -T1Q1;
- Rb and Rc are each independently a protecting group or -T1Q1, or Rb and Rc, together with the nitrogen atom to which they are attached, form an unsubstituted or substituted heterocycle comprising one or two 5- or 6-member rings and 1-4 additional heteroatoms selected from N, O and S;
- T1 is a bond or unsubstituted or substituted C1-C6 alkyl linker;
- and
- Q1 is H, hydroxyl, halogen, unsubstituted or substituted C1-C6 alkyl, unsubstituted or substituted C1-C6 alkoxy, unsubstituted or substituted 2-6 membered heteroalkyl, unsubstituted or substituted C3-C10 carbocycle, or unsubstituted or substituted heterocycle comprising one or two 3-, 4-, 5- or 6-member rings and 1-4 heteroatoms selected from N, O and S,
- or a pharmaceutically acceptable salt, ester, solvate, or hydrate thereof,
- wherein said treating ototoxicity comprises reducing cell death or increasing cell survival of a cell from the subject suffering from ototoxicity.
