Abstract
This invention is directed toward a non-wild-type organophosphorus acid anhydrolase enzyme having two site mutations, method of production, and method of use to more effectively degrade toxic organophosphorus compounds and, in particular, toxic chemical GD (3,3-Dimethylbutan-2-ylmethylphosphonofluoridate), than the wild type organophosphorus acid anhydrolase.
Claims20
An isolated organophosphorus acid anhydrolase (OPAA), wherein said anhydrolase comprises a non-wild-type amino acid at each of sequence positions 212 and 342 of SEQ ID NO:
- 1.
A method for degrading GD (3,3-Dimethylbutan-2-ylmethylphosphonofluoridate), comprising contacting GD with an isolated organophosphorus acid anhydrolase, wherein said anhydrolase comprises a non-wild-type amino acid at sequence positions 212 and 342 of SEQ ID NO:
- 1.
A kit for degrading GD, comprising:
- (a) an isolated organophosphorus acid anhydrolase, wherein said anhydrolase comprises a non-wild-type amino acid at sequence positions 212 and 342 of SEQ ID NO:
- 1;
- and
- (b) a pharmaceutically-acceptable carrier.
