Abstract
Isolated aspartyl-tRNA synthetase polypeptides and polynucleotides having non-canonical biological activities are provided, as well as compositions and methods related thereto.

A pharmaceutical composition, comprising a physiologically-acceptable carrier and an aspartyl-tRNA synthetase (AspRS) polypeptide of up to 200 amino acids in length that comprises (a) an amino acid sequence that is at least 95% identical to residues 1-154, 1-171, or 1-174 of SEQ ID NO:1, or (b) an active fragment of (a) which is 140 or more contiguous amino acids of (a), wherein the AspRS polypeptide or active fragment thereof comprises an OB-fold domain and an amphiphilic helix, wherein the polypeptide has a non-canonical activity selected from induction of IL-10 and binding to TLR2, TLR4, or both, and wherein the composition has an LPS-containing bacterial endotoxin level of less than about 12 EU/mg.
