## Abstract

The invention relates to vaccine compositions including CEV serogroup immunogens, attenuated and inactivated viruses of the CEV serogroup and chimeric Bunyaviridae. Also disclosed are methods of treating or preventing CEV serogroup infection in a mammalian host, methods of producing a subunit vaccine composition or an immunogenic composition, isolated polynucleotides comprising a nucleotide sequence encoding a CEV serogroup immunogen, methods for detecting La Crosse virus (LACY) infection in a biological sample and infectious chimeric Bunyaviridae.

## Claims

### Claim 1

An immunogenic composition comprising a La Crosse virus polypeptide or a nucleic acid molecule encoding same, wherein said La Crosse virus polypeptide comprises a mutation at amino acid 148 of GN.

### Claim 2 (depends on 1)

The composition of claim 1, wherein the mutated amino acid at position 148 of GN is Alanine.

### Claim 3 (depends on 1)

The composition of claim 1, wherein amino acid 148 of GN is deleted.

### Claim 4 (depends on 1)

The composition of claim 1, wherein the mutated amino acid at position 148 of GN is any amino acid other than Threonine.

### Claim 5 (depends on 1)

The composition of claim 1, wherein said LACV polypeptide is a LACV/human/1960 virus sequence, LACV/mosquito/1978 virus sequence, or a LACV/human/1978 virus sequence.

### Claim 6 (depends on 5)

The composition of claim 5, wherein said LACV polypeptide sequence has at least about 85%, 88%, 90%, 92%, 95%, 97%, 98%, 99%, or 99.9% amino acid identity to an amino acid sequence of SEQ ID NOs. 7, 8, 9, 10, 11 or 12.

### Claim 7 (depends on 1)

The composition of claim 1, wherein said nucleic acid molecule is codon-optimized for expression in humans.

### Claim 8 (depends on 1)

A method of inducing an immune response against an LACV infection in a mammal, comprising administering to said mammal a therapeutically effective amount of the composition of claim 1.

### Claim 9 (depends on 1)

A method of producing an immunogenic composition for inducing an immune response against an LACV, comprising the steps of providing the composition of claim 1; and formulating said composition for delivery to a human.
