## Abstract

Certain embodiments described herein are directed to chromatography systems that include a microfluidic device configured to provide three-way switching or switching between three or more inputs or outputs. The microfluidic device can be fluidically coupled to one or more switching valves to provide for selective control of fluid flow in the chromatography system.

## Claims

### Claim 1

A method of modulating flow of a fluid in a chromatography system including a microfluidic device comprising a laminated wafer body comprising an internal microchannel fluidically coupled to a first port and a second port, wherein the microfluidic device is fluidically coupled to a first switching valve through the first port of the microfluidic device and to a second switching valve through the second port of the microfluidic device, the method comprising:
independently actuating the first switching valve and the second switching valve between a first position and a second position, the first position of the first switching valve permitting fluid flow from a switching gas source to the first port of the microfluidic device, the first position of the second switching valve permitting fluid flow from the gas source to the second port of the microfluidic device, in which the actuation of the first switching valve and the second switching valve is configured to provide three-way switching using the microfluidic device; and
altering fluid pressure to permit a sample to flow to a first outlet port of the microfluidic device in a first state of the microfluidic device, in which the first outlet port is fluidically coupled to the first port and the second port through the internal microchannel, and to permit the sample to flow to a second outlet port of the microfluidic device in a second state of the microfluidic device, in which the second outlet port is fluidically coupled to the first port and the second port through the internal microchannel.

### Claim 2 (depends on 1)

The method of claim 1, in which each of the first and second switching valves is a 3-way solenoid valve.

### Claim 3 (depends on 2)

The method of claim 2, further comprising balancing pressure in the system by configuring the microfluidic device with at least one restrictor bypass flow path.

### Claim 4 (depends on 2)

The method of claim 2, further comprising configuring the microfluidic device with a third, fourth, fifth and sixth port, in which the first switching valve is fluidically coupled to the switching gas source, the first port and the third port, and the second switching valve is fluidically coupled to the switching gas source, the third port and the fifth port.

### Claim 5 (depends on 4)

The method of claim 4, further comprising configuring each of the first and second switching valves to be in the first position to provide switching gas to the first port and the fifth port and to provide column effluent to the second port.

### Claim 6 (depends on 4)

The method of claim 4, further comprising configuring the first switching valve to be in the second position and the second switching valve to be in the first position to provide switching gas to the third port and the fifth port and to provide column effluent to the sixth port.

### Claim 7 (depends on 4)

The method of claim 4, further comprising configuring the first switching valve to be in the first position and the second switching valve to be in the second position to provide switching gas to the first port and the third port and to provide column effluent to the fourth port.

### Claim 8 (depends on 4)

The method of claim 4, further comprising configuring each of the first and second switching valves to be in the second position to provide switching gas to the third port and to provide column effluent to the fourth port and the sixth port.

### Claim 9

A system comprising:
a microfluidic device comprising a laminated wafer body comprising an inlet port, a first outlet port and a second outlet port, in which each of the first outlet port and the second outlet port is fluidically coupled to the inlet port through an internal microchannel in the laminated wafer body, the microfluidic device constructed and arranged to permit three-way switching;
a first switching valve fluidically coupled to the microfluidic device and configured to provide a switching gas to at least two ports of the microfluidic device to provide sample flow from the inlet port to the first outlet port; and
a second switching valve fluidically coupled to the microfluidic device and configured to provide a switching gas to at least two ports of the microfluidic device to provide sample flow from the inlet port to the second outlet port.
