Abstract
This document describes biochemical pathways for producing pimelic acid, 7-aminoheptanoate, 7-hydroxyheptanoate, heptamethylenediamine, or 1,7-heptanediol by forming two terminal functional groups, comprised of carboxyl, amine or hydroxyl group, in a C7 aliphatic backbone substrate produced from chorismate or benzoate. These pathways, metabolic engineering and cultivation strategies described herein rely on the anaerobic benzoyl-CoA degradation pathway enzymes.
Claims31
A method for biosynthesizing 7-aminoheptanoate, said method comprising, in a recombinant host:
- enzymatically synthesizing a seven carbon chain aliphatic backbone from benzoyl-CoA and
- enzymatically converting the seven carbon chain aliphatic backbone to 7-aminoheptanoate using:
- a thioesterase classified in EC 3.1.2-, a dehydrogenase classified under 1.2.1-, a CoA-transferase classified in EC 2.8.3.12, or a CoA-ligase classified in EC 6.2.1.5 to form a terminal carboxyl group, and
- a ω-transaminase classified in EC 2.6.1—to form a terminal amine group, wherein the host is a bacteria, yeast, or plant cell.
A recombinant host comprising at least one exogenous nucleic acid encoding (i) a chorismate lyase or an anthranilate synthase;
- (ii) a benzoyl-CoA reductase and (iii) a 2-ketocyclohexane-1-carboxyl-CoA hydrolase, said host producing pimeloyl-CoA.
A method for biosynthesizing 7-aminoheptanoate, said method comprising:
- enzymatically synthesizing a seven carbon chain aliphatic backbone from benzoyl-CoA and
- enzymatically converting the seven carbon chain aliphatic backbone to 7-aminoheptanoate using:
- a polypeptide having the activity of a thioesterase classified in EC 3.1.2-, a dehydrogenase classified under 1.2.1-, a CoA-transferase classified in EC 2.8.3.12, or a CoA-ligase classified in EC 6.2.1.5 to form a terminal carboxyl group, and/or
- a ω-transaminase classified in EC 2.6.1-to form a terminal amine group, wherein the method is performed in vitro.
