Abstract
The invention is directed toward non-wild-type organophosphorus acid anhydrolases having three site mutations, method of production, and method of use to effectively degrade toxic chemical compounds such as (Ethyl({2-[bis(propan-2-yl)amino]ethyl}sulfanyl)(methyl)phosphinate (“VX”).
Claims9
An isolated organophosphorus acid arthydrolase (OPAA), wherein said anhydrolase comprises a non-wild-type amino acid at sequence positions 212, 215 and 342 of SEQ ID NO:
- 1, and wherein tyrosine (Y) is replaced by phenylalanine (F) at position 212, isoleucine (I) is replaced by phenylalanine (F) at position 215, and valine (V) is replaced by leucine (L) at position 342.
A method for degrading (Ethyl({2-[bis(propan-2-yl)amino]ethyl}sulfanyl)(methyl)phosphinate) (VX), comprising:
- contacting VX with an isolated organophosphorus acid anhydrolase, wherein said anhydrolase comprises a non-wild-type amino acid at sequence positions 212, 215 and 342 of SEQ ID NO:
- 1, and wherein tyrosine (Y) is replaced by phenylalanine (F) at position 212, isoleucine (I) is replaced by phenylalanine (F) at position 215, and valine (V) is replaced by leucine (L) at position 342.
A kit for degrading (Ethyl({2-[bis(propan-2-yl)amino]ethyl}sulfanyl)(methyl)phosphinate) (VX), comprising:
- an isolated organophosphorus acid anhydrolase, wherein said anhydrolase comprises a non-wild-type amino acid at sequence positions 212, 215 and 342 of SEQ ID NO:
- 1, and wherein tyrosine (Y) is replaced by phenylalanine (F) at position 212, isoleucine (I) is replaced by phenylalanine (F) at position 215, and valine (V) is replaced by leucine (L) at position 342;
- and
- a pharmaceutically-acceptable carrier.
