Abstract
Isolated human monoclonal antibodies which bind to human CD74 and related antibody-drug conjugates are disclosed. Pharmaceutical compositions comprising the antibodies or antibody-drug conjugates, and therapeutic and diagnostic methods for using the antibodies and/or antibody-drug conjugates, are also disclosed.

An antibody which binds to CD74 variants 1 and 2 and comprises a VL region comprising the CDR1, 2 and 3 sequences set forth in SEQ ID NO:24, SEQ ID NO:27, and SEQ ID NO:25, respectively, and
- a) a VH region comprising the CDR1, 2 and 3 sequences set forth in SEQ ID NO:
- 20, 21 and 22, respectively;
- b) a VH region comprising the CDR1, 2 and 3 sequences set forth in SEQ ID NOS:8, 9 and 10, respectively;
- c) a VH region comprising the CDR1, 2 and 3 sequences set forth in SEQ ID NO:
- 12, 13 and 14, respectively;
- or
- d) a VH region comprising the CDR1, 2 and 3 sequences set forth in SEQ ID NO:
- 16, 17 and 18, respectively.
An antibody which binds to CD74 variants 1 and 2 and comprises:
- (a) a VH region comprising the sequence of SEQ ID NO:
- 19 and a VL region comprising the sequence of SEQ ID NO:
- 26;
- (b) a VH region comprising the sequence of SEQ ID NO:
- 7 and a VL region comprising the sequence of SEQ ID NO:
- 26;
- (c) a VH region comprising the sequence of SEQ ID NO:
- 7 and a VL region comprising the sequence of SEQ ID NO:
- 23;
- (d) a VH region comprising the sequence of SEQ ID NO:
- 11 and a VL region comprising the sequence of SEQ ID NO:
- 26;
- or
- (d) a VH region comprising the sequence of SEQ ID NO:
- 15 and a VL region comprising the sequence of SEQ ID NO:
- 26.
A multispecific antibody, comprising a first antigen-binding region of an antibody selected from the group consisting of:
- (a) an antibody comprising a VH region comprising the sequence of SEQ ID NO:19 and a VL region comprising the sequence of SEQ ID NO:26;
- (b) an antibody comprising a VH region comprising the sequence of SEQ ID NO:7 and a VL region comprising the sequence of SEQ ID NO:23;
- (c) an antibody comprising a VH region comprising the sequence of SEQ ID NO:11 and a VL region comprising the sequence of SEQ ID NO:26;
- and
- (d) an antibody comprising a VH region comprising the sequence of SEQ ID NO:15 and a VL region comprising the sequence of SEQ ID NO:26, and
- at least one second antigen-binding region having a different binding specificity.
An expression vector comprising a nucleotide sequence encoding one or more of the amino acid sequences selected from the group consisting of SEQ ID NOs:
- 7, 11, 15, 19, and 23, or any combination thereof.
