Abstract
Methods for making a biodegradable collagen matrix having increased osteoinductivity and a biodegradable collagen matrix prepared by these methods are provided. In various embodiments, the methods include providing an acidic collagen slurry and mixing it with at least one water soluble and/or hydrophilic bioactive agent under conditions sufficient to cause the collagen slurry to self-assemble into macroscopic collagen fibers and cause the at least one bioactive agent to form a collagen matrix containing the bioactive agent. Conditions sufficient to cause the collagen slurry to self-assembly include raising the pH of the slurry to from about 5 to about a pH of 9 and/or adding bone powder, calcium phosphate, hydroxyapatite, DBM or a mixture thereof to the acidic collagen slurry in order to raise the pH from about 5 to about 9.
Claims12
A method for making a biodegradable collagen matrix, the method comprising:
- providing an acidic collagen slurry;
- mixing at least one water soluble and/or hydrophilic bioactive agent with the acidic collagen slurry under conditions sufficient to cause the collagen slurry to self-assemble into macroscopic collagen fibers having a length from about 3 mm to about 30 mm, from about 5 mm to about 25 mm or from about 7.5 mm to about 20 mm and cause the at least one bioactive agent to form a collagen matrix containing the bioactive agent;
- adding an osteoclastogenesis inhibitor to the collagen matrix;
- and
- adding an osteoinductive additive comprising particulate demineralized bone matrix comprising bone chips,
- wherein the conditions sufficient to cause the collagen slurry to self-assemble comprise:
- (i) raising the pH of the slurry from 8 to about a pH of 9;
- or
- (ii) adding bone powder, calcium phosphate, hydroxyapatite, DBM or a mixture thereof to the slurry to raise the pH from 8 to about 9, and
- disposing the formed collagen matrix into a containment device comprising a biodegradable porous polymer mesh bag.
A method for making a biodegradable collagen matrix, the method comprising:
- providing an acidic collagen slurry;
- adding a dried mixture of BMP-2 and hydroxyapatite to the acidic collagen slurry under conditions sufficient to cause the collagen slurry to self-assemble into macroscopic collagen fibers having a length from about 3 mm to about 30 mm, from about 5 mm to about 25 mm or from about 7.5 mm to about 20 mm and causing the BMP-2 to form a collagen matrix containing the BMP-2;
- adding an osteoclastogenesis inhibitor to the collagen matrix;
- and
- adding an osteoinductive additive comprising particulate demineralized bone matrix comprising bone chips,
- wherein the conditions sufficient to cause the collagen slurry to self-assemble comprise:
- (i) raising the pH of the slurry from 8 to about a pH of 9;
- or
- (ii) adding the hydroxyapatite to the slurry to raise the pH from 8 to about 9, and
- disposing the formed collagen matrix into a containment device comprising a biodegradable porous polymer mesh bag.
