A pharmaceutical composition, comprising an isolated modified mRNA polynucleotide that encodes an aminoacyl-tRNA synthetase (AARS) polypeptide that is at least 95% identical to SEQ ID NO:20 or 32, or a fragment thereof which is at least 300 contiguous amino acids of SEQ ID NO:20 or 32, wherein the AARS polypeptide has an extracellular signaling activity and wherein the composition is substantially endotoxin free.
