Abstract
In one aspect, the invention relates to compounds having the formula: where R1, R2, R3, X, R4, R5, and R6 are as defined in the specification, or a pharmaceutically acceptable salt thereof. These compounds have neprilysin inhibition activity. In another aspect, the invention relates to pharmaceutical compositions comprising such compounds; methods of using such compounds; and process and intermediates for preparing such compounds.

A compound of formula I:
- where:
- R2 is OH;
- R3 is —CF3;
- R1 is —OR10;
- R10 is H or —C1-6alkyl;
- X is pyridyltriazole;
- R4 is H;
- R5 is H;
- and R6 is H.
(2R,3R)-4-(2-chloro-phenyl)-2-hydroxy-3-[(3-hydroxy-isoxazole-5-carbonyl)-amino]-butyric acid.
(2S,3R)-3-[(7-chloro-3H-benzotriazole-5-carbonyl)-amino]-4-(2-chloro-phenyl)-2-hydroxy-butyric acid.
(R)-4-(2-chloro-phenyl)-3-[(2-hydroxy-pyrimidine-5-carbonyl)-amino]-butyric acid.
