Abstract
To provide a more convenient and more accurate method of assaying ProGRP by improving the stability of ProGRP which is known to be unstable in a biological sample. By using a blood sample in a condition in which a blood coagulation factor is not activated is used as a sample, the degradation of ProGRP is suppressed, whereby it is possible to store a sample for a long period of time and to improve the accuracy of an assay.
Claims2
A method of assaying pro-gastrin-releasing peptide (ProGRP) in a serum sample, the method comprising:
- (a) obtaining the serum sample from a subject;
- (b) adding a serine protease inhibitor to the serum sample to form a mixture of the serine protease inhibitor and the serum sample;
- (c) storing the mixture at room temperature, wherein more than 80% of ProGRP 31-98 region present in the mixture after step (b) remains in the mixture after 20 hours of storage at room temperature;
- and
- (d) assaying the mixture for ProGRP 31-98 region following the storing step (c),
- wherein the serum sample is not dried, the serine protease inhibitor is added to the serum sample at time 0, and the serine protease inhibitor is phenylmethylsulfonyl fluoride.
