Provided herein are Nα,Nα,Nα-trialkyl histidine derivative compounds and methods of their preparation. Also provided are methods of their use for preparing useful compounds such as ergothioneine.

A method of producing a compound of Formula 3:
- comprising:
- (a) reacting a compound of Formula 1 with a compound of Formula 2 to form the compound of Formula 3:
- wherein each R1 is (C1-4)alkyl.
A method of producing a compound of Formula 5:
- comprising:
- (c) reacting a compound of Formula 4 with a thiocyanate to form the compound of Formula 5:
- wherein each R1 is (C1-4)alkyl.
