Abstract
This document describes biochemical pathways for producing butadiene by forming two vinyl groups in a butadiene synthesis substrate. These pathways described herein rely on enzymes such as mevalonate diphosphate decarboxylase, isoprene synthase, and dehydratases for the final enzymatic step.
Claims35
A method for the biosynthesis of butadiene comprising
- converting a butanol or butene to 3-buten-2-ol using (a) a polypeptide having the activity of a desaturase or (b) using a polypeptide having the activity of a monooxygenase to form a first terminal vinyl group, wherein said desaturase or monooxygenase forms a first vinyl group, and wherein said desaturase is a cytochrome P450 or said monooxygenase is a gene product of mdpJ, and
- forming a second terminal vinyl group in the 3-buten-2-ol using a polypeptide having the activity of a dehydratase in enzyme class EC 4.2.1 capable of accepting a hydroxylated substrate to biosynthesize butadiene.
The method of 5, wherein the principal carbon source fed to the fermentation derives from biological or non-biological feedstocks.
The method of 7, wherein the biological feedstock is or derives from monosaccharides, disaccharides, lignocellulose, hemicellulose, cellulose, lignin, agricultural waste or municipal waste.
